Heterologous expression in E. coli

The bacterial expression system is the most used and more economic resource to obtain heterologous proteins. BiCT is able to offer a fast and economic feasibility service for the production of amount of protein, initially for research use or for the first screening of activity and selectivity.
Given the industrial nature of the research in BiCT, the company also offers a service of strain improvement and optimization of fermentation and purification conditions for achieving production targets according to customer needs, building a path leading to the transfer of a process to industrial scale.

 

Heterologous expression in yeast

The yeasts are a heterologous expression system industrially relevant. BiCT has the know-how to offer customers the ability to perform the feasibility of processes in different hosts: Saccharomyces cerevisiae, Kluyveromyces lactis and Pichia pastoris.
It is also possible, in case of positive feasibility, to proceed with the improvement of the expression and strain, up to industrial targets defined together with the customer.

 

In development: heterologous expression in fungi (Aspergillus)

The filamentous fungi represent an extremely efficient heterologous expression system, with consolidated industrial applications for the production of enzymes. BiCT is developing internally an expression system based on the micro-organism Aspergillus, which will become strategic for the production of a broad category of enzymes.

The use of natural or recombinant strains has a common denominator represented by cell cultures, in order to obtain the appropriate amount of biomass or related product. BiCT has a wide competence in microbiological field, both in terms of operational scale (microplates, flasks and fermenters) and type of cells: from bacteria, E. coli up to the most complex Actinomycetes, up to yeasts, filamentous fungi and cell lines less conventional, such as plant cells.

 

BiCT can provide its experience in various operational stages:

  • screening and optimization of the growth conditions
  • development of the scale up
  • strain improvement (increased productivity)
  • microbiological and chemical-physical analysis
  • definition of the strain preservation conditions

The obtainment of a particular product very often entails the development of a purification method that allows to recover the molecule with defined parameters and an appropriate purity profile. BiCT is able to develop and optimize purification processes by exploiting the most suitable technologies, by the filtrations dead-end or tangential to the most different chromatography techniques: ion exchange, gel-filtration, hydrophobic interaction, mixed mode, affinity and reverse phase with the stationary phases most suitable for each specific application. In the specific case of chromatography, BiCT can simultaneously develop the process and become a supplier of the resin / matrix on an industrial production scale.

BiCT offers to its customers its expertise in the field of enzyme immobilization, by adopting the most appropriate carrier and the most suitable technique (adsorption, covalent binding, modification of the spacer arm, etc.).

 

We immobilize for you:

  • One or more commercial enzymes currently used in your free-form processes
  • An enzyme of your property

If the enzyme of interest is not commercially available and needs to be developed, BiCT with its expertise in the field of the expression of heterologous proteins as well as the subsequent purification, can provide it for feasibility studies.

 

BiCT, thanks to its experience in both the biotechnology and chemical field can develop and optimize biocatalytic processes mediated by enzymes, or multisteps reactions with chemo-enzymatic steps. The enzymes can be used either free or immobilized, depending on the application and convenience.
The use of immobilized enzymes at the industrial level can provide the following advantages:

  • reduce procurement costs of bio-catalysts, due to the possible of enzyme reuse
  • in some fields, such as food, not to haveresidual enzyme
  • allow theuse of enzyme even in the presence of solvents, as it occurs in the solid phase non-aqueous
  • improve the stabilityof a catalyst
  • confer particular characteristics of reaction or modify the kinetic parameters of the biocatalyst
